چکیده

Background—Many cells that migrate during normal embryonic development or in metastatic cancer first detach from an epithelium. However, this step is often difficult to observe directly in vivo and the mechanisms controlling the ability of cells to leave the epithelium are poorly understood. In addition, once cells detach, they must assume a migratory phenotype, involving changes in cytoskeletal and signaling dynamics. Drosophila border cells provide a model system in which forward genetics and live cell imaging can be combined to investigate the cellular and molecular mechanisms of epithelial cell detachment and migration in vivo.

Par-1 Controls Myosin-II Activity through Myosin Phosphatase to Regulate Border Cell Migration

BACKGROUND Localized actomyosin contraction couples with actin polymerization and cell-matrix adhesion to regulate cell protrusions and retract trailing edges of migrating cells. Although many cells migrate in collective groups during tissue morphogenesis, mechanisms that coordinate actomyosin dynamics in collective cell migration are poorly understood. Migration of Drosophila border cells, a g...

Complete Polarization of Single Intestinal Epithelial Cells upon Activation of LKB1 by STRAD

The LKB1 gene encodes a serine/threonine kinase that is mutated in the Peutz-Jeghers cancer syndrome. LKB1 is homologous to the Par-4 polarity genes in C. elegans and D. melanogaster. We have previously reported the identification and characterization of an LKB1-specific adaptor protein, STRAD, which activates LKB1 and translocates it from nucleus to cytoplasm. We have now constructed intestina...

Mammalian homologues of C. elegans PAR-1 are asymmetrically localized in epithelial cells and may influence their polarity

The establishment of polarity in the embryo is fundamental for the correct development of an organism [1]. The first cleavage of the Caenorhabditis elegans embryo is asymmetric with certain cytoplasmic components being distributed unequally between the daughter cells [2-4]. Using a genetic screen, Kemphues and co-workers have identified six par genes (partition-defective) [5,6], which are invol...

Neural crest specification by noncanonical Wnt signaling and PAR-1.

Neural crest (NC) cells are multipotent progenitors that form at the neural plate border, undergo epithelial-mesenchymal transition and migrate to diverse locations in vertebrate embryos to give rise to many cell types. Multiple signaling factors, including Wnt proteins, operate during early embryonic development to induce the NC cell fate. Whereas the requirement for the Wnt/β-catenin pathway ...
